Peran Nrf2 Dalam Patogenesis Stres Oksidatif dan Inflamasi pada Penyakit Ginjal Kronik

Abstract


Penyakit ginjal kronik (PGK) merupakan masalah kesehatan serius yang dapat membatasi usia harapan hidup dan meningkatkan biaya kesehatan. PGK ditandai dengan penurunan fungsi dan kerusakan struktur ginjal serta dapat mengalami komplikasi kardiovaskular ataupun berkembang menjadi penyakit ginjal stadium akhir. Stres oksidatif dan inflamasi diketahui berperan dalam patogenesis PGK. Ketidakseimbangan reactive oxygen spesies (ROS) dan pertahanan antioksidan mencetuskan keadaan ini. Nuclear factor-erythroid-2 related factor 2 (Nrf2) merupakan faktor tanskripsi yang dapat meningkatkan transkripsi berbagai antioksidan dan enzim-enzim detoksifikasi. Gen-gen protektif ini akan dengan cepat menetralkan ROS. Akan tetapi, pada PGK terjadi gangguan aktivasi dari Nrf2.
